Abstract
The objective of this study is to describe a novel dry powder insufflator (DPI) developed for the delivery of powder aerosols to a small animal model (rat). In vitro and in vivo performance evaluation of the DPI will be described and a series of formulation applications will be presented. The basic components of the air-jet DPI for rat delivery (Rat AJ DPI) are an air source, air-jet aerosolization engine and delivery cannula with safety wedge. In order to benchmark the new device, we compared the in vitro and in vivo aerosol performance of the Rat AJ DPI with the Penn-Century DP-4 DPI (which is no longer commercially available) using a model albuterol spray dried formulation at different delivered doses. We also evaluated the in vitro and in vivo performance of the Rat AJ DPI using other spray dried formulations (tobramycin and a lung surfactant) to explore its ability to deliver a variety of dry powder aerosols. The Rat AJ DPI demonstrated efficient emptying and small-particle aerosol generation making it suitable for preclinical powder formulation evaluation in rat animal models.
